Abstract

High-performance thin layer chromatography combined with in vivo evaluation offers a robust framework for validating herbal hepatoprotectives such as Tinospora cordifolia. In the present work, an ethanolic stem extract was standardized through phytochemical screening and High-performance thin layer chromatography fingerprinting, then assessed against paracetamol-induced liver injury in Wistar rats. Authenticated plant material was extracted, chemically profiled, and administered orally at 200 and 400 mg/kg for seven days. Biochemical markers and histopathology were evaluated, with silymarin serving as reference. The extract revealed abundant alkaloids, diterpenoid lactones, flavonoids, and phenolics, while High-performance thin layer chromatography produced reproducible bands corresponding to key marker compounds, confirming quality and identity. Paracetamol markedly elevated liver enzymes and disrupted hepatic architecture; treatment with T. cordifolia significantly restored biochemical balance and preserved tissue structure in a dose-dependent manner, approaching silymarin at the higher dose. Protective effects appear related to antioxidant reinforcement, membrane stabilization, and modulation of inflammatory responses. The integrative approach links chemical fingerprints with biological performance, strengthening the scientific basis for standardization, authentication, and therapeutic consistency of herbal preparations. Findings highlight the value of High-performance thin layer chromatography as a quality-control platform and support further exploration of T. cordifolia as a safe, economical hepatoprotective option. Future efforts should isolate bioactive markers, clarify molecular mechanisms, and investigate long-term safety to enable rational formulation and potential clinical translation. Such evidence can guide regulatory acceptance, encourage batch-to-batch reproducibility, inform dose selection, and ultimately bridge traditional knowledge with modern, evidence-based hepatology for broader patient access and sustained therapeutic reliability across global settings.
